功能描述

SUMO结合增强子。通过与UBE2I/UBC9相互作用,通过促进E1和E2酶的结合、SUMO与UBE2I/UBC9之间的硫酯键连接以及SUMO向特定靶蛋白(包括HIF1A、PIAS、NFKBIA、NR3C1和TOP1)的转移,增加蛋白质的SUMO结合。Isoform 1和isoform 2通过增强NF-kappa-B抑制因子alpha (NFKBIA)的SUMO化,正向调节NF-kappa-B信号通路,促进其稳定,进而导致对NF-kappa-B转录活性的抑制增加。Isoform 1和isoform 2通过增加HIF1A的SUMO化,负向调节缺氧诱导因子-1 alpha (HIF1A)信号通路,在缺氧期间促进其稳定、转录活性及其靶基因VEGFA的表达。Isoform 2促进糖皮质激素受体NR3C1的SUMO化和转录活性,并增强SUMO1和NR3C1与UBE2I/UBC9的相互作用。对泛素化无影响。 {ECO:0000269|PubMed:17956732, ECO:0000269|PubMed:22009797, ECO:0000269|PubMed:23469069, ECO:0000269|PubMed:23508108}。

组织特异性

Isoform 1 and isoform 2 are expressed in glioma tumors (at protein level). Expressed in a wide number of tissues with highest expression in cerebellum, pituitary, heart, kidney, liver, stomach, pancreas, prostate and spleen. Low levels in thalamus, spinal cord, esophagus, thymus, lung and peripheral blood leukocytes. A higher level expression seen in pituitary tumors as compared to the pituitary gland.
