功能描述

既可作为NAD(P)H依赖性还原酶,又可作为S-亚硝酰-CoA依赖性亚硝酰转移酶的Enzyme (PubMed:10620517, PubMed:18241201, PubMed:27207795, PubMed:38056462, PubMed:7929092)。在发育过程中促进胎儿血红素降解 (PubMed:10858451, PubMed:18241201, PubMed:7929092)。也在成体组织中表达,作为造血、中间代谢(谷氨酰胺分解、糖酵解、TCA循环和磷酸戊糖途径)及胰岛素信号传导的调节因子发挥作用 (PubMed:27207795, PubMed:29500232, PubMed:38056462)。具有广泛的底物特异性氧化还原酶活性,通过催化多种黄素(如核黄素、FAD或FMN)、胆绿素、高铁血红蛋白和PQQ(吡咯喹啉醌)的NAD(P)H依赖性还原发挥作用 (PubMed:10620517, PubMed:18241201, PubMed:7929092)。通过在肝脏中催化胆绿素IXbeta还原为胆红素IXbeta,促进胎儿血红素分解代谢 (PubMed:10858451, PubMed:18241201, PubMed:7929092)。胆绿素IXbeta是胎儿体内主要的血红素分解产物,在成体中不存在 (PubMed:10858451, PubMed:18241201, PubMed:7929092)。不能还原胆红素IXalpha (PubMed:10858451, PubMed:18241201, PubMed:7929092)。在FMN和NADPH存在下,也能将络合的Fe(3+)铁还原为Fe(2+) (PubMed:10620517)。作为蛋白质亚硝酰转移酶,催化靶蛋白(如HMOX2、INSR和IRS1)半胱氨酸残基的亚硝基化 (PubMed:38056462)。S-亚硝酰-CoA依赖性亚硝酰转移酶活性通过“乒乓”机制介导:BLVRB首先与S-亚硝酰-CoA和蛋白质底物结合,然后一氧化氮基团从S-亚硝酰-CoA转移到BLVRB的Cys-109和Cys-188残基,再从S-亚硝酰-BLVRB转移到蛋白质底物 (PubMed:38056462)。通过介导INSR和IRS1的亚硝基化抑制胰岛素信号传导,导致其受抑制 (PubMed:38056462)。 {ECO:0000269|PubMed:10620517, ECO:0000269|PubMed:10858451, ECO:0000269|PubMed:18241201, ECO:0000269|PubMed:27207795, ECO:0000269|PubMed:29500232, ECO:0000269|PubMed:38056462, ECO:0000269|PubMed:7929092}。

Predominantly expressed in liver and erythrocytes (PubMed:7929092). At lower levels in heart, lung, adrenal gland and cerebrum (PubMed:7929092). Expressed in adult red blood cells (PubMed:29932944).
